Petri film Plates Preparation Instructions

 
Sample Preparation
Cut Sample to small pieces Put them into plastic bag and weighted
Fill 90 ml distil water in the sample bag Blend on chopping board with hammer

Aerobic Count Plate (A/C)

Inoculation
With pipette perpendicular to AC, place 1 ml of sample onto center of bottom film

Inoculation
Gently apply pressure on spreader to distribute inoculums over circular area before gel is formed. Do not twist of slide spreader.
Incubation
Incubate plates with clear slide up in stacks of no more than 20.
Interpretation
After 24 hour, AC can count with illuminated magnifier.

E. Coli/Coli form Count Plate (EC)
Inoculation
With pipette perpendicular to EC, place 1 ml of sample onto center of bottom film
Inoculation
Gently apply pressure on spreader to distribute inoculums over circular area before gel is formed. Do not twist of slide spreader.
Incubation
Incubate plates with clear slide up in stacks of no more than 20.
Interpretation
After 24 hour, Coli forms can count and after 48 hours EC can count with illuminated magnifier.

With pipette perpendicular to RSA, place 1 ml of sample onto center of bottom film.
